Back in grad school some 25 years ago, we mixed colchicine with agar and let it gel in a petri dish. Then we stuck the cotyledons into the gel for the required time. We left the roots up in the air, or else turned the dish over on its cover so the roots would at least hang down. though the roots were in the air, the ari had a humidity of about 100%.
We were crossing wheat, rye, and barley in all combinations.
