If you've got a copy of Howard Shockey's article, would you consider
sending it out to all of us along with the basic "shopping list" (what
power microscope, etc.)?
<
No. You can order a copy of the 1991 YB itself from ASI for only six
dollars -- think of it as only 50 cents a page for Howard's comprehensive
instructions and all the other articles are "free".
The microscope specs are easy, though. You need a quality instrument
[roughly 1000x] with good eyepieces, an oil immersion objective, and
built-in substage illumination. [Howard's article went into more detail,
but those are the basic requirements. It should be an instrument designed
for photography -- but certainly doesn't have to be a fancy electron
microscope.
My 600x scope didn't quite cut it. Good enough to determine ploidy. Not
good enough to isolate marker chromosomes and far too awkward for
photography. But it still gets a workout on pollen analysis.
