If Roundup isn't working it either isn't being used properly or there's a part of the puzzle missing. Used at the right concentration Roundup should kill both annual and perennial weeds though some times a second application is necessary. There are also other, stronger herbicides that will essentially sterilize the area but you need to be careful that non-target weeds and ornamentals are not effected. Once the Roundup had done it's job I'd think you could apply a pre emergent in a granular form to prevent additional weeds from germinating...corn gluten might work or a traditional chemical treatment. If the weed seeds are coming in from adjacent planted areas then it's going to be an ongoing battle that needs a battle plan.
