This is a very good question.  I know of no one in the US now doing chromosome
counts on iris.  ASI has a member in Germany who has done counts within the last
few years.  There is an article by Howard Shockey concerning technique in the
Aril Society International yearbook for 1991 (p 40).  Howard did a
number of iris chromosome counts over the years. He references some older 
books and articles on technique.  There is at least one recent book about chromosome
counting technique (I don't have the reference at hand), but it doesn't specifically
discuss iris and is aimed more at professional plant scientists.
Chromosome counting isn't something that can be taken up casually with any
expectation of success.  

A chromosome count of course would be the most direct way of proving a
Japanese-Siberian hybrid.

I will be delighted to learn that someone is doing
counts, so anyone knowing of such an individual please respond.
