You need to dig this out, including as much underground
root as you can manage...that ought to do it in.

If you just pull runners without taking care of the source, they will
simply come back, generally in multiples as where it breaks off a
larger runner, it seems to initiate more than one growth bud.
This is true - if you don't get the source, you'll simply have it back. Rather than spray the entire plant however, follow the various creeping bits back to where they root (you might find multiple rooting sections on a major plant) Cut the vines off from the roots at each rooted point and then paint the freshly cut trunk of the root with *undiluted* Roundup. Ready to spray mixtures are too weak for this application. Make sure you follow it to its source to get the "mother" root. Cut the branches off and paint this as well with the undiluted Roundup.

Roundup is registered for this use. This is the single best way to remove unwanted vines, weedy trees etc without indiscriminate spraying.

Naturally, you'll wear long rubber gloves and take all other recommended safety precautions on the label. :-)

Any living or thriving vines in the area after two weeks means you missed painting and cutting a root.
