The vine has turned a yellowish color instead of a rich green like the other
plants.  Most likely Genitic and nothing to worry about.

The secondary vines stopped at about two inches, and their leaves stopped
growing. There doesn't
seem to be any insect activity or signs of chewed leaves, it just sorta
stopped growing.

Could this be a fungus, bacteria, or maybe the soil is missing some
nutrient?   Are you watering regularly ?  Did you over or under fertilize ?

P.S. Can an Atlantic Giant be used for making pumpkin pie, or is it just
grown for size?
They are just grown for size. The pumpkins most used for pumpkin pies are " Sugar Pumpkins".
