Not any experience in counting Iris, but one thing comes to mind.
Counts made on meiotic material are usually much more accurate than from somatic
tissue (particularly from root tips). Root tips are notorious for having
irregularities, due usually apparently to the rapid growth with tends to lead to
mistakes. A common mistake is the failure of a cell to split after the
chromosomes replicate, leading to false polyploid counts. On the other
hand, some groups are apparently less prone to these problems than others, and
from what I recall (somebody correct me if I'm wrong), root tips seem to be the
standard for Iris counts.
