What happens when you apply colchine is that during cell division, a cell
wall fails to form as the microtubers do not form properly during
chromosome doubling and alignment in the center of the cell. A spindle
apparatus does not form and the cell forms with double the chromosome
number. Basically, you will be turning AG from a diploid plant to a
tetraploid plant. They say that it will enlarge fruit and the like ,
however, some have found that it has strange effects on fruit size making
it smaller sometimes. I dont know what happens to the pumpkin genome, but
there are also cases of mutations. Also, I think a colchicine application
works better once the plant has germinated.
