Rick, There are 2 universitys within 40 miles. Wash. St. U. and U of
Idaho. I majored in Plant Sciences at the U of Idaho. I also worked at
the U of I as a research tech. for a few yrs in another life. To be honest
I have no idea what is happening to the genetic material after I treat with
cholchicine. I run my treatments in groups of 10 seeds, soaking them for
24hrs. At the same time I place 10 of the same seeds in water for 24hrs.
This helps establish germination rate and helps determine the concentration
I need for effect. The treated seeds don't develope as fast in the seed
and germination takes a day or two longer. If the untreated germinate 100%
and the treated 10% I know my solution is too strong. If treated and
untreated are both 100% and germination time is the same I know its too
weak. Each time I start a new treatment I also start an untreated check.
I germinate in damp papertowles in ziplocks, both the treated and untreated
are placed in the same general area with respect to heat and light. They
both are aired several times daily, at the same time. When the roots begin
to elongate, I move the seedling to peat pots. This is the first time that
the effect of the treatments may be visible. At this point if there is no
difference I begin again. As the treated begin to grow the differences
from the untreated become more apparant. Yet with all of this there is no
clue as to the size of the Pumpkin that may result. Have fun, Roger
