No, the pumpkin cavity is sealed from the stem.

The stem is hollow as is the vine.  However, cut the
stem from the pumpkin close to the shell and it turns
solid at that point.  If it did not, when you cut a
pumpkin from the vine you would have a hole into the
cavity and it would rot very quickly.
